Walkthrough:

Buy a space bike post and place it somewhere outside your base to complete goal #3.

The previously forbidden zone to the north of the base opens up. (The barrier of red lights changes to green, and you can now build and explore there.) Buy and place some lights in the new zone to uncover the marker beacon. That's goal #1.

Buy a weeding post and place it that northern zone among the podulus toxicaria and lupulus weeds. Set Stig on weeding duty, since he has the highest skill level in Weeding. Have him burn the podulus toxicaria first, otherwise it will release mutant bees, which can harm your operatives and your base structures, while seeding additional weed plants.

An operative on weeding duty will work his way outward from the weeding post. To make him burn weeds in a certain spot, first assign him to the weeding post. When he has picked up the flamethrower, click on him, then click on the area where you want him to start burning. Green arrows indicate the spot where he'll start.

Check the Base Conditions report on the Bridge Screen. When you succeed in getting Vegetation Encroachment levels down to 0%, you've met goal #2.

Other tips: With an additional operative available, you'll have a little more flexibility in assigning jobs. For example, if you have enough base nutrients, set Venus on power (she has 2 stars) and have Dean and Stig do weeding. Have Tami alternate between cleaning and oxygen.

Again, new equipment becomes available at the start of the mission, including exercise equipment for Venus and Dean.
